چکیده

Introduction: Glycosylation is one of the post-translational modifications on proteins and lipids. Change in glycosylation on the cancer cell surface could be associated with malignancy characteristic such as invasion and metastasis. Most breast cancer deaths occurs in due to the invasion of tumor cells to vital organs such as lung, liver, brain. One of the molecules related to aberrant glycosylation is sialic acids (SAs) .St6galnac5 gene (siat7e ), encodes a golgi type II transmembrane glycosyltransferase that plays an important role in cellular adhesion and involved in breast cancer metastasis to brain. These enzymes catalyze transfer of sialic acid from CMP-Neu5Ac to the glycan moiety of glycol conjugate on the cell surface. Material and methods: Total RNA was extracted from breast cancer cell line . cDNA was provided and used as a template in RT-PCR. Finally products were purified and were cloned into pGEM-T vector. white colonies were screened for the presence of the correct insert by pcr and enzymatic digestion and finally was sent for sequencing. Results : Extracted sequences cloned into pGEM-T vector for sequencing. The extracted plasmids were sequenced and aligned with different siat7e sequences. The results revealed about more than 99% identity with sequences that are presented in Genbank. Conclusion: Altered sialylation is important in cancer transformation and metastasis. Siat7e encodes a protein that is responsible for the synthesis of disialoganglioside that involved in cell adhesion. Recent studies have showed that this protein act as a mediator (decrease cell adhesion) in metastatic cancer cell line So we can look for the ways to prevent cancer metastasis by more understanding of sialylation. In this study Two pairs of primers were designed based on the sialyl motifs . This motifs plays main role in sialylation of glycoproteins.
