Introduction: Breast cancer is the most common cancer in woman around the world.Like other carcinomas, breast cancer occurs based on an interaction between geneticheterogeneity and the environment. An accumulation of genetic variants is involved inthe process of breast carcinogenesis. Among these genetic variants, many of them playroles in apoptosis. Apoptosis is a programmed cell death. The Inhibition of Apoptosisplays an important role in the development of malignancy. The most important antiapoptoticprotein is Bcl2. Bcl2 can protect cells against apoptosis induced by agentswith different mechanism of action and can increase resistance to anticancer drugs.Objective: Bioinformatic studies of Bcl2 gene show a region with CA sequencerepeats in the promoter of Bcl2 gene. To date, there has been no study on thepolymorphism and the association of this microsatellite with breast cancer. The aim ofthis study is to analyze the CA polymorphism in the promoter region of Bcl2 gene andto evaluate its association with breast cancer in Esfahanian patients.Material and methods: Genomic DNA was extracted from the blood of patients withbreast cancer and matched control and after designing primers, the promoter of Bcl2gene was amplified by PCR. Finally the length of the repetition sequence is analyzedby polyacrylamide gel.Results: So far we have observed alleles with different number of CA repeats in thepromoter region of Bcl2 gene in breast cancer patients and its relationship with breastcancer is under investigation.Conclusion: Characterizing the number of CA repeats in the promoter region of thisgene and understanding its relationship with breast cancer can help identifyindividuals predisposed to breast cancer and can also have therapeutic purposes.
