Study of immune responses of new variety of Brucella melitensis in BALB/c mice

Brucellosis is one of the most widespread zoonosis in the world affecting many domestic and wild animals including bovines, goats ,sheep, pigs and dogs. Although effective, the current Brucella vaccines (Rev.1 and M5-90) have several drawbacks. So in the present study immunological and protective abilities of a new variety of Rev-1 vaccine strain of Brucella melitensis was evaluated in BALB/C mice. Thirty mice were divided randomly to three groups each consisted of ten mice. Three groups of mice were immunized respectively with Rev-1 ,new variety (Pars variety ) and 3th group as control received cell culture media. The humoral immune responses were estimated by ELISA and protective immunity was evaluated by challenging the all groups with lethal dose of Brucella melitensis. IgG was elevated in mice receiving Rev-1 and it’s variant , while IgG2a was elevated in mice receiving varient of Rev-1. All the immunized mice were protected against challenge with infective dose of Brucella, while mice of control group were died during two weeks post challenge.
