Effect of amino acids and precursors on the growth of recombinant CHO cells

In this study, the effect of five amino acids and nucleic acids as precursors on the growth of recombinant CHO (rCHO) cells was investigated. The rCHO cells were cultivated in basal medium composing DMEM/F12 (1:1 v/v) and fetal bovine serum (10%) and 262×105 cells/ml was obtained. The results showed that threonine, asparagine, methionine increased the rCHO cells growth up to 35%, 20% and 24% in comparison with basal medium, respectively. But the effect of two other amino acids, proline and glycine, on the cell growth was not considerable. Nucleic acids (ATP, GTP, CTP and TTP) as main precursors increased the rCHO cells growth significantly (42%). The ammonium concentration was daily measured as a toxic by-product. It was not increased in presence of amino acids. On the other hand, the rCHO cells grew at higher densities with adding amino acids compared to the basal medium. It seems that the positive effect of amino acids on the cell growth is not related to their effect on ammonium concentration. The ammonium was formed by adding the precursors in lower amounts compared with the basal medium. The results indicated that the precursors increased the cell growth more than amino acids. Therefore, there is a reverse relation between ammonium concentration and cell growth. In summary, the medium enriching with amino acids and/or precursors is suggested as a suitable medium for growth of rCHO cells.
