Association of p53 Intron1 Polymorphism and Prostate Cancer Risk

چکیده
The tumor suppressor gene p53 is thought to play major roles in the development of humanmalignancy. Polymorphic variants of p53 have been found to be associated with cancersusceptibility, but few studies have investigated their effect on prostate cancer risk. In thiscase-control study, we investigated the correlation of TAAAA repeat polymorphisms in p53intron1 with prostate cancer risk. To our knowledge this is the first report about polymorphicTAAAA repeat of the P53 gene and its relation to prostate cancer risk. A total of 260peripheral blood samples consisting of 130 from patients with prostate cancer and 130 fromhealthy control individuals were included in the study. Total genomic DNA was extracted bymeans of standard techniques. The DNA samples were genotyped using PCR amplificationwith specific primers to TAAAA repeat sequence of p53 intron1. The PCR products wereresolved on a polyacrylamide gel and visualized by silver nitrate staining. The exact size ofthe alleles was determined by sequencing the PCR products. Five different alleles havebeen found for polymorphic TAAAA repeat of p53 gene in our study population. The shortestand the longest alleles contained 6 and 10 repeats of TAAAA, respectively and allele with 8-TAAAA repeat had the most frequency in both patients and control individuals. Preliminaryresults show that the distribution of the major genotypes in patients and controls is different.Preliminary findings of this study show that pentanucleotide repeat in the intron 1 of p53 genemay be associated with prostate cancer risk. So the results could help identify people at riskof prostate cancer.کلیدواژه ها
