We have assessed the effect of feeding different levels of copper-methinione treatment onascites incidence and MMP-2 changes in lungs of broiler chickens reared under lowtemperature conditions. Growing the broiler chickens in cold temperature can induce ascites.A total of 480 broiler chickens were studied within 6 treatments ( two ambient temperatureand there levels (0, 100, 200 mg/kg of diet) of copper- methionine ) and four replication.Following 4 wk of rearing at normal ambient temperatures, half of broilers were exposed tolow temperature (15-19 ÌŠC ) and half of them were kept in normal temperature (25-28 ÌŠC)from 28 days to 45 days. Ascites were detected at 45 d of age in broilers reared in coldtemperature without feeding copper- methionine according to abdominal and pericardial fluidaccumulation. Four broilers of each treatment were selected at days 45 of age. The lungswere collected, and immediately stored at -80 ÌŠC for gelatin zymography, gelatin reversezymography, real-time PCR and enzyme activity assay. The data indicated that the matrixmetalloproteinases-2 (MMP-2) activity increased in lung of ascitis broilers. The results ofzymography demonstrated that feeding with copper-methionine significantly decreased pro-MMP-2 in cold condition. Pro-MMP-2 gene expression was the highest in lung of ascitisbroiler. Tissue inhibitor matrix metalloproteinases-2 (TIMP-2) didnt change in lungs ofbroilers reared in different treatments. These results suggest that feeding copper-methioninedecreased MMP-2 and ascites possibility in broilers reared in cold temperature, thereforefeeding with copper-methionine as a cofactor for superoxide dismutase ( antioxidant enzyme)may be useful for prevention of lung cancer.
