Alpha gliadin is the most abundant wheat seed proteins. Gluten is classified in two families of glutenin and gliadin proteins .Gliadins are divided in three structural classes of α, ɣ and ωgroups [2] Celiac disease is also known as gluten enteropathy, a form of gluten intolerance and is an inherited disorder of the immune system that damages the lining of the small intestine. It is caused by gluten consumption; however, α-gliadin components are mostimportant to allergens. One of the effective methods to control allergen gens expression is RNAi technology (RNA interference) that is used to extinguish or reduce gene expression. The purpose of this research is to identify the structure of α-gliadin and producingproprietary RNAi cassette for wheat α-gliadin. So that firstly nucleotide sequence of α- gliadin gene involved in designed for PCR amplification by online primer-blast software. The amplificant were extracted from agarose gel and ligated to pTG19 cloning vector. After cloning of the recombinant plasmid in E.coli, they were sequenced. Following verification of DNA sequence, they were used for construction of RNAi cassette. The constructed RNAicassette containing two inverted repeat fragments of α-gliadin gene with a 150 bps fragment in between. This plasmid is used for silencing specific genes and provided a means to control allergen genes and reduce the gluten.
