Albumin based nanoparticles have gained considerable attention as drug delivery carriers as they hold advantages suchas being non-toxic. Cationic human serum albumin nanoparticles (HSA NPs) can be used as carriers of negatively chargeddrugs and simultaneously to increase the cellular uptake. Three strategies were employed for the preparation of cationicHSA NPs using polyethylenimine (PEI): (I) coating HSA NPs with PEI polymer through attractive electrostaticinteractions, (II) coating HSA NPs with PEI via covalent amide bond formation between amine groups of PEI andcarboxyl groups of HSA NPS, and (III) coating HSA NPs with PEI via covalent amine-amine bond formation betweenamine groups of PEI and amine groups of HSA NPS. Among the above examined strategies, strategy II was moreeffective to coverage HSA NPs with PEI and resulted in a higher shift in the zeta potential value of HSA NPs towardpositive values. Finally, gallic acid was used as a negatively charged molecule and loaded into PEI-HSA NPs.
