چکیده مقاله:

Transcription factors (TFs) regulate direct reprogramming of mouse fibroblast to cardiomyocyte . These TFs interact with other proteins to form protein complexes during regulation of gene expression. Our study focus on predicting the most important protein complex in protein-protein interaction network during conversion of mouse fibroblast to cardiomyocyte.Method: Data of GSE 22292 accession number was downloaded from GEO database. Normalization of raw data were done using Affymetrix Powerful Tool (APT) and differential expressed (DE) genes identified using fold change algorithm. ChIPEnrichment Analysis OF DE genes reveal TFs binding sites. Agilent literature search plugin of cytoscape and BioGRIDwere used to find protetin interactions for TFs. Cytoscape was used for visualization of network. Protein complexes of network find using MCODE plugins in cytoscape.Result: Our analysis show 1522 DE genes for conversion of mouse fibroblast into cardiomyocyte. We identify 25 TFs the have arole in regulation of most of these DE genes. TFs protein-protein interaction network contain 144 nodes with 278 interactions. MCODE analysis of this network show protein complex (highest score complex 4.25) with 9 nodes and 19 edges. This protein complex contain SUZ12, EZH2, SFPI1, IKZF1, II7R, LY6A, IGF1, NPPB, and NPPA that play roleduring direct reprogramming of mouse fibroblast to cardiomyocyte. Interestingly SUZ12, EZH2, and SFPI1 are TFs involvein regulation of this conversion and present in most important protein complex. Conclusion:TFs which contribute in this protein complexes may be have an important role in regulation of mouse fibroblast conversion into cardiomyocyte.
