چکیده مقاله:

Xylanase is employed as an ingredient in animal feed to improve digestibility and enhancing the efficacy of nutrient absorption by facilitating the conversion of hemicellulose, originally locked in the cell walls of the grains, to sugars. As such the nutrients are easier to be released and the animal can then obtain better energy with less feed. Cost effectiveness of feed is thus attained. Since the Trichoderma fungus has the ability to produce enzymes like Xylanase which has the effectiveness of breaking down hemicellulose and xylan component of the cell wall of the plant, producing the livestock and poultry food can be possible by applying this ability to agricultural waste. In this study, mutation induction in T. harzianum with optimum dose of ۲۵۰ Gy based on appearance ۴۰-۵۰% of the spore germination was selected and after substrate preparation and induction of xylanase enzyme mutant isolates activity of ۲۰ mutants have been evaluated. gamma–irradiated Trichoderma harzianum are collected from the surrounding soil of sugar beet root fields in East Iran, and held in vitro cultured PDA. TCM and TFM are used for the growth of spores and inducing the production of xylanase. Finally the enzyme is measured at ۵۰ºC for ۶۰ minutes by DNS method and the samples are absorbed by spectrophotometry at wavelength ۵۴۰ nm.and the amount of xylanase enzymes produced from gamma–irradiated Trichoderma harzianum mutants is measured.
