Background: Foot-and-mouth disease is an important transboundary animal disease with a significant economic impact. One of the important factors in the effectiveness of the Foot-and-Mouth disease vaccine in preventing and eradicating the disease is the efficiency and potency of the vaccine, which is directly related to the virus titer. Therefore, one of the concerns in producing a inactivated Foot-and-Mouth disease virus vaccine is obtaining a higher viral titer. Methods: BHK-۲۱ monolayer cells were exposed to cold shock of ۱۰°C for ۲۰, ۴۰, and ۶۰ minutes, and heat shock of ۴۰ and ۴۲°C for ۱۰, ۲۰, and ۳۰ minutes, and combined shocks of first cold of ۱۰°C and then heat of ۴۰ and ۴۲°C according to the previous times. Then, Foot-and-Mouth disease virus serotype O was added to it. After observing ۸۰% CPE, the virus titer was examined and measured using the Reed and Munch method, and the results were compared. Results: In the combined cold shock of ۱۰°C for one hour and heat shock of ۴۲°C for ۲۰ minutes, an increase in viral titer of ۱ logarithm was observed. In other models of cold shock alone, heat shock alone, or combined cold and heat shock, no significant change in viral titer was observed. Conclusion: Given the role of HSP in increasing the attachment of the foot-and-mouth disease virus to the cell and facilitating the process of virus entry into the cell, as well as increasing the viral transcription genes in cold and heat shocks on BHK۲۱ cells and increasing the titer of the foot-and-mouth disease virus produced for the purpose of inactivated vaccines, a larger dose of vaccine can be produced with a smaller volume of virus.
