Chronic wounds represent a significant and growing clinical challenge, primarily due to the establishment of tolerant bacterial biofilms. Within these biofilms, Quorum Sensing (QS) serves as the master regulatory system coordinating communal behaviors such as virulence factor production and extracellular polymeric substance (EPS) synthesis. This review synthesizes contemporary evidence (۲۰۱۵-۲۰۲۵) on strategies to inhibit QS, a process known as Quorum Quenching (QQ). It details the mechanisms of major QS systems in key wound pathogens, evaluates the efficacy of natural and synthetic QS inhibitors (QSIs), and explores their translational applications in medicine, agriculture, and industry. While QSIs show considerable promise in disrupting biofilms and attenuating pathogenicity without exerting direct bactericidal pressure, challenges such as emerging resistance, delivery optimization, and the need for robust clinical validation remain. The future of this field lies in developing integrated, multi-targeted anti-virulence therapies and smart delivery systems to translate QQ strategies into effective clinical tools for managing chronic wounds and combating antimicrobial resistance.
