Evaluation of WRKY Transcription factor۱ Expression in Cumin ( Cuminum cyminum) Plants under Drought Stress and Silicon Nanoparticle Foliar Application

Cuminum cyminum, a valuable medicinal plant from the Apiaceae family. Drought stress significantly affects its yield and the production of secondary metabolites. WRKY transcription factors play a crucial role in these defense mechanisms. The application of nanosilica has emerged as a modern approach to mitigate the effects of drought stress in medicinal plants. This research was conducted to assess the extent and type of changes in the expression of the WRKY TF۱ gene in this plant. The Isfahan ecotype was used for semi-quantitative analysis. The experiment was carried out in a randomized block design. Drought stress treatments included ۶۰%, ۴۰%, and ۲۰% of field capacity, and silicon nanoparticle treatments were applied at three levels: ۰ (control), ۴, and ۶ mM. Following primer design, RNA extraction, cDNA synthesis, and gene fragment amplification were performed using a thermal cycler. The results of the ۱.۵% TAE agarose gel were analyzed using Image Lab software. The results indicated that drought stress affected the expression of the WRKY gene in Cuminum cyminum. Additionally, foliar application of silicon nanoparticles also influenced this expression pattern, with the highest gene expression observed at ۴۰% field capacity drought stress and ۴ mM silicon nanoparticle concentration.
