Callus Induction, Shoot and Root Regeneration in Hyssopus officinalis Using Sodium Nitroprusside and Plant Growth Regulators

The current study aims to assess the callus induction and regeneration of shoots and roots in Hyssopus officinalis L. using sodium nitroprusside (SNP) and plant growth regulators (PGRs). A significant impact was found in the callogenesis of leaf and petiole explants with different concentrations of SNP and ۲,۴-dichlorophenoxyacetic acid (۲,۴-D). SNP concentrations of ۲۰ µm induced callogenesis to ۷۲%, but when combined with ۲,۴-D concentrations, callogenesis increased to ۱۰۰%. In the absence of SNP, no calluses were developed. A significant effect of ۲,۴-D and SNP was observed on callus fresh weight. Callus fresh weight from leaf explants was ۰.۱۶۵ g, while from petiole explants was ۰.۱۳ g. Furthermore, SNP enhanced shoot regeneration of leaf explants compared to petiole explants. When ۲۰ µm of SNP was combined with ۳ mg/L ۶-benzylaminopurine (BAP), the maximum mean shoot number was obtained (۸۴% in leaves and ۵۴% in petioles). A combination of SNP and IBA enhanced rooting rate to ۸۳% and root length to ۸.۳ cm. After acclimation, over ۸۰% of the produced plants survived in greenhouse conditions. Several implications can be drawn from the study's findings regarding medicinal hyssop cultivation, the preservation of genetic resources, and pharmaceutical research on medicinal plants.
