In the present study a combined analytical method involving ultrasonic extraction (USE) and headspace liquid phase micro extraction (HS- LPME) was used to extract methyl tert-butyl ether (MTBE) in fish tissue for analysis by gas chromatography (GC). Extraction of MTBE from ۲ g of fresh fish tissue was performed by ultrasonication for ۱۰ min (in ۲۵ ºC after ۳.۵ h maceration) in the glass barrel that was filled with water and sealed. Headspace solvent microextraction was done for the extraction of MTBE after centrifuge of sonicated sample. The optimized method (under the optimal experimental conditions: ultrasonic extraction time: ۱۰ min, ultrasonic extraction temperature: ۲۵ °C, fish particle size: a particle of ۲ grams, media: distilled water, maceration time: ۳.۵ h and centrifuge of sonicated sample: ۲۵۰۰ rpm in ۱۵ ˚C for ۱۰ min) had good linearity (R۲ >۰.۹۹) over the range ۰.۵- ۵۰ µg g-۱, and showed satisfactory level of precision, with RSD values in the range of ۱.۱ to ۹.۷ %. The LOD about ۰.۲ µg g-۱ was obtained.
