Expression Profile of HOXB ۶-۱۳ Genes in Cumulus Cells of Women with Polycystic Ovaries

چکیده مقاله:

Background: ۱۰-۱۵ percent of women in fertility age sufferfrom polycystic ovarian syndrome (PCOS). PCOS is awoman-related disease that is recognized by existence two ofthe three criteria: hyperandrogenism, oligo anovulatory andpolycystic ovary, that ۲۰% of normal female population mayshow Polycystic ovary disease (PCOD) in ultrasound definition,which have normal hormone concentration and menstrualcycle. Genetic disorder is one of the most important PCOS’sfactors affecting oocyte and it’s around tissue such as cumuluscells during follicogenesis. Matter of HOX genes as mastergenes in cell function and development is studied in many dis eases. In this research, the expression profile of HOXB۶-۱۳genes in the cumulus cells of women with PCOS and PCODwere focused.Materials and Methods: The study groups were included of۲۲ PCOS and ۸ PCOD versus ۱۰ normal women. After RNAextraction and cDNA synthesis, mRNA expression of the candidategenes was measured by real-time PCR.Results: Data showed increased expression of HOXB۶(P=۰.۰۰۲), HOXB۷ (P=۰.۰۰۲), HOXB۸ (P=۰.۰۲), andHOXB۱۳ (P=۰.۰۰۱) in cumulus cells of women with PCOS vs.control. However, there were no significant changes in mRNAlevels of the aforementioned genes in PCOD group vs. controland nor PCOS.Conclusion: This study give us an idea about a significant correlationbetween altered expression of HOXB members andPCOS disorder, and implies the dynamic role of these developmentalgenes in pathogenesis of polycystic ovary symptoms.
