Troxerutin Affects Histological Structure of Ovary in Offspring of High Fat-Induced Obese Rats by Reduction of Apelin-۱۳ Receptor Expression

Background: Feeding and physical condition of mother canaffect offsprings development. Previous studies indicated thatobesity and high fat diet (HFD) feeding during pregnancy havedeleterious cosequences on reproductive functions of the offsprings.It is also indicated that apelin and its receptor play acritical role in human follicular development and pathogenesisof polycystic ovary syndrom. Troxerutin (TX) is a naturalbioflavonoid which has many biological effects and improvesobesity and its related metabolic parameters in HFD-treatedmice. This study was conducted to evaluate the effects of theTX administration during pregnancy on histological structureof ovary, plasma level of apelin-۱۳, and expression of its receptorin ovarian tissue of HFD fed rats offsprings.Materials and Methods: In this study, ۴۰ female ۳ week-oldwistar rats were randomly divided into the normal and HFDgroups. Normal group received standard diet while HFD groupreceived HFD diet for ۸ weeks and then placed with normalmale rats for copulation. After mating, pregnant animals ofeach group were devided into two subgroups including teratedand non-treated with TX (۱۵۰ mg/kg/day) by oral gavage untilthe end of pregnancy. Femal offsprings of all groups selectedand fed normal diet. Ninty days after birth (PND۹۰), ۳۲ offspringseuthanized and their blood samples and ovarian tissuewere collected to serum concentration of apelin-۱۳, histologicalstructure of ovary and expression of apelin-۱۳ receptor inovarian tissue be evaluated. Serum concentration of apelin-۱۳detected by ELISA kit. Histological and histomorphometricalstudies were carried out on paraffin embedding tissue sectionand H&E stained slides and expression of apelin-۱۳ receptorsin ovary tissue analyzed by real-time PCR. Finally, data werestatistically analyzed by SPSS using one-way ANOVA test andTukey's post-hoc (α=۰.۰۵).Results: Results showed that HFD reduced serum concentrationof apelin-۱۳ in comparison with control groups significantly(P<۰.۰۵) and TX had not significant effect (P>۰.۰۵).Ral-time PCR analysis indicated that HFD incresaed expressionof apelin-۱۳ receptors in ovary tissue and TX could reduceit significantly (P<۰.۰۱). Microscopical studies revealed thatHFD decreased percent of primary, secondary and graph folliclesand icreased atretic follicles significantly (P<۰.۰۵) whileTX administration could improved this structures in the level ofcontrol groups.Conclusion: Based on our results it can be concluded thatdministration of TX in high fat diet fed mothers during pregnancycan be effective as a suitable supplement to improvestructural changes of ovary in their offsprings by reduction ofapelin-۱۳ receptor.
