The Effect of Various Concentrations of Eggplant Peel Extract on Quality Membrane Integrity Farahani RamSperm Breed

Background: This study was conducted by investigating theeffect of different concentration eggplant peel extract (۰, ۲, ۴, ۶and ۸ percent/ml) in extender with based Tris-egg yolk on theexperiment Farahani ram breed. Semen samples were collectedfrom five mature Farahani ram breed twice in the week by anartificial vagina, after equilibration at five degrees centigradethe samples, sealing by ۰.۵ straw and were frozen with nitrogenvapor and immersed in liquid nitrogen and were stored byusing for assessment. Semen’s were thawed at ۳۷ºC and thenassessed for analysis of membrane integrity (HOST) by computeranalysis (CASA). The result of this experiment showedthat the extender containing about ۲ percent/ml of eggplantpeel extract significantly improved cell membrane integritycompared to control group (P<۰.۰۱). After that, the extenderscontaining about ۴ percent/ml from eggplant extract peel andcontrol groups had an equal relationship of significant (P<۰.۰۱).With the attention, this conclude of this experiment addition ۲percent/ml of eggplant peel extract to Tris extender could besuitable for preservation of cell membrane integrity.Materials and Methods: Semen samples were obtained fromfive Farahani ram around (۲-۳ years old) and the average weightof ۵۰-۶۰ kg. This experiment was carried out in October andDecember at experimental station of Arak University. Semenwas collected with an artificial vagina at two times a week. Bythe way, for prevention of individual effects, the semen sampleswere polled and the ejaculations were immediately transferredto a water bath with ۳۷ºfor transportation to the laboratory. Thevalue of every ejaculation was about ۱-۲ ml and concentration۲/۵×۱۰۹ spermatozoa/ml healthy and normal spermatozoa.Results: The results of this study showed that extender restrainsof ۲ percent/ml eggplant peel extract could be decreased cellmembrane integrity compared to control group after freezethawing,although the level of ۴ percent/ml and control group atthis experimentation had equal of statistical significance in cellmembrane integrity (P<۰/۰۱).Conclusion: In this experiment, our results showed that using ۲percent/ of eggplant peel extract in the condition of the laboratorycould improve membrane integrity and quality of spermatozoaafter the freeze. However, based on the results acquiredin the present study greater level addition did not grant moreuseful effect.
