Methanolic Extract of Coconut Meat Decrease Apoptosis of Testis by Improvement of Oxidative Stress in Type II Diabetic Rats

Background: The increasing incidence of diabetes mellitus(DM) and its complications has made this disease one of thebiggest health threats to the communities. There is a strong associationbetween male infertility and DM and about ۹۰% ofmale diabetic patients have abnormalities in the fertility. Coconutis one of the highest nutritional and medicinal value plantsand our previous studies showed that methanolic extract of coconutmeat (MECM) can improve quality and quantity indicesof sperm in diabetic rats. This study was conducted to evaluatethe effect of MECM on apoptosis and oxidative stress in testisof type II diabetic rats.Materials and Methods: Twenty-five adult male Wistar ratswere divided into ۵ groups including control, Diabetic and ۳treated diabetic groups which received ۱۰۰, ۱۵۰ and ۲۰۰ mg/kg/day MECM by oral gavage for ۴۰ consecutive days. TypeII diabetes was induced by high fat diet and ۳۵ mg/kg streptozotocin.Finally, animals were euthanized and their left testiswas removed. Tissue samples were processed by routine andstandard paraffin embedding and sectioned. Detection of apoptoticcell was performed by nonradioactive in situ end labelingmethod using TUNEL immunocytochemical technique. A partof testis tissue were homogenized and level of glutathione peroxidase(GPx), superoxide dismutase (SOD), malondialdehyde(MDA) and total antioxidant capacity (TAC) were measured.Finally, data were statistically analyzed by SPSS using one-wayANOVA test and Tukey's post-hoc (α=۰.۰۵).Results: The results showed that induction of diabetes significantlyincreased Apoptotic indices and MDA level of testisbut decreased GPx, SOD and TAC level of testis while in alltreatment groups, these conditions were improved in a dosedependent manner. Our results indicated that administrationof MECM in all three dose reduced apoptotic indices andMDA level of testis and also increased GPx, SOD and TAClevel of testis tissue significantly compared to diabetic groups(P<۰.۰۱). There is no significant difference between mid andhigh dose of MECM groups and control groups in these regards(P>۰.۰۵).Conclusion: Based on our results it can be concluded that administrationof MECM can decrease apoptosis in diabetic patientsby reduction of oxidative stress in testis tissues.
