Background: Chemotherapy is one of the risk factors whichaffect the fertility. Paclitaxel (PTX) is an effective chemotherapeuticagent and mitotic inhibitor which binds to DNA and preventsthe DNA replication. PTX is used against a wide rangeof solid tumors. It has been reported numerous functional andstructural alterations in testicular tissue following the administrationof PTX. Monosodium glutamate (MSG) is the L-form ofglutamic acid. Some studies indicated that monosodium glutamatecan induce oxidative stress and has toxic effects on humanand animal’s tissues. Male infertility and alteration of spermproduction and morphology are the most reported changes incases of MSG administration. In this study the effects of coadministrationof PTX and MSG on the microscopic structure ofseminiferous tubules (STs) was evaluated.Materials and Methods: Paclitaxel (۲ mg/kg i.p.) was administeredto mice once per day for ۵ consecutive days. MSG (۳۰& ۶۰ mg/kg i.p.) was administrated for ۲۸ days before or afterPTX administration. Formaldehyde fixed testicular tissue sampleswere stained with hematoxylin and eosin method for quantitativeevaluation of STs.Results: The mean diameter of STs was reduced significantly inall groups in comparison to control group. Coadministration ofPTX and MSG dose dependently led to more reduction in STsdiameter. Tubular atrophy was observed in more degree followingthe administration of MSG after PTX.Conclusion: Structural changes in testicular STs, followingthe administration of PTX can be related to fertility alterations.Oxidative compounds such as MSG can induce more structuraland functional alterations of testicular tissue in cases of chemotherapies.
