Background: The use of adipose tissue derived mesenchymalstem cells (ADMSCs) for the treatment of central nervoussystem diseases and anti cancer drug delivery in animal andclinical human researches has been attention during the recentyears. The limitation of stem cell migration to the brain is oneof the most important barriers to effective treatment with stemcells. Therefore, many studies have been done to understand themechanisms and functions of genes which related to stem cellmigration into the brain. In previous studies, the effect of leukemiainhibitora factor (LIF) on the potency of stem cells,variousstages of neurogenesis and repair of brain and spinal cord injuryhave been shown. The purpose of the present study was toevaluate the effect of over expression of LIF in ADMSCs cellson the gene expression of adhesion molecules and cytokine receptorsrelate to the transmission of stem cells from the bloodbrainbarrier.Materials and Methods: ADMSCs was expanded from pretesticularadipose tissue of male C۵۷BL/۶ mice based on digestionwith collagenase I and serial passage in DMEM mediumcontaining ۱۵% FCS and antibiotics. Isolated cells were phe notypically characterized based on determination of expressionof OCT۴ and NANOG genes by conventional PCR, presenceor absence of some surface markers including CD۹۰, CD۴۴,CD۳۱ & CD۴۵. In the present study ADMSCs was transfectedby Eukaryotic pIRES۲-EGFP vector containing LIF gene usingcalcium-phosphate method. Expression of CXCR-۱, CXCR-۴,CCR-۲, LFA-۱, MAC-۱, VLA-۴ & PSGL-۱ and was analyzedusing real time PCR at days۷ and ۱۴ after transfection. Nontransfected ADMSCs were used as negative control.Results: Our results showed that CXCR-۱, CXCR-۴, CCR-۲,LFA-۱, MAC-۱, VLA-۴ were transcribed in ADMSCs, whilePSGL۱ had no expression in ADMSCs. Over expression of LIFin ADMSCs resulted in elevation of expression of all studiedadhesion molecules and cytokine receptors, in particular, ۱۴days after transfection.Conclusion: Given the stimulatory effects of LIF overexpressionon the transcription of adhesion molecules and cytokinereceptors genes in ADMSCs, It is likely that this method can beused to improve the transmission of stem cells for treatment ofneurodegenerative diseases in future.
