Background: There has been an explosion of research, publicationsand conference in the field of stem cells in the lastdecade. Mesenchymal stem cells (MSCs) are undifferentiatedcells found in many adult tissues, including adipose, bone marrow,brain, muscle, ocular, heart, liver etc. In addition, MSCshave been considered as an appropriate source for gene andcell therapy tools for tissue engineering and regenerative medicine.To clarify different aspects of MSCs biology and developMSCs applications in human diseases treatment, other studieson animal models are required. However, the similarity betweenzebrafish and human genome has been further examined;among other animal models that have been used, zebrafish isconsidered to be the best for practical purposes in regenerativemedicine. In this report, multi-potency and pluri-potency potentialof heart and liver derived-MSCs was presented for the firsttime. The aim of this study was to investigate the multipotencyand pluripotency of zebrafish heart and liver derived-MSCs forusing zebrafish as suitable animal model for regenerative medicinein cell therapy.Materials and Methods: In the present study, MSCs were isolatedfrom heart and liver tissue of Zebrafish (Danio rerio) aspreviously reported in another papers. The flow-cytometry aswell as RT-PCR was used to analyze the expression of a panelof cell surface markers CD۴۴, CD۹۰, CD۳۱ and CD۳۴. In thefollowing, alizarin red, oil Red-O and toluidine blue stainingwere carried out to evaluate the multipotency of zebrafish heartand liver tissue-derived MSCs. Subsequently, the gene expressionsof Oct۴, Sox۲ and Nanog as pluri-potent markers wereanalyzed by RT-PCR.Results: The results showed that, like other MSCs, zebrafishheart and liver tissue-derived MSCs were expressed pluripotentmarkers Oct۴, Sox۲ and Nanog. Moreover, these cells weredifferentiated to osteocyte, adipocyte and chondrocyte lineagesfollowing directed differentiation. However, the findings of thisstudy are consistent with the hypothesis that MSCs have similarfeatures with other MSCs in different tissue.Conclusion: Our results show that both heart and liver tissuehave similar characteristics including multi-lineage differentiationcapacity and pluripotency potential.
