Background: Heart failure remains a leading cause of mortalityin the World. Reduction in myocyte number and cardiacpump dysfunction are marked in many of the heart failure suchas myocardial infarction. So, find a method to recover cardiomyocytefunction after myocardial injury or to increase thenumber of cardiomyocytes, it has been a long-term goal. Use ofstem cells which can be transplanted into the infarcted myocardiumthat can repair and regenerate the lost tissue and improvecardiac function, as an attractive method has been reported. Theuse of C-kit+ cardiac stem cells (CSCs) as an appropriate candidatecell for future cardiac regenerative medicine strategiesis promising. C-kit+ CSCs have been extensively examined,consistently exhibit several in vitro characteristics that definea “stem cell”, being clonogenic, self-renewing and multipotentpotential. But the cellular mechanisms of C-kit CSCs have yetto be reported. The aim of this study is to determine the effectof C- kit+ CSCs co-culture on angiogenic factors of cardiomyocytesderived adult rat heart via VEGF signaling pathway.Materials and Methods: C-kit+ stem cells were isolated frombone marrow of adult rattus norvegicus by magnetic cell sorting(MACS) (Miltenyi), and the purity of the isolation was assessedby FACS analysis. Subsequently, cardiomyocytes wereisolated from adult rat heart by enzymatic digestion and bothcell types were co-cultured with each other for ۴۸ hours. In thefollowing, cardiomyocytes were collected; quantitative RealtimePCR and western blotting were used for the detection ofgene expression of GATA۴ and TBX۵ and protein expression ofVEGF, respectively.Results: The results of this study showed that the expression ofGATA۴ and TBX۵ genes and VEGF protein was significantlyincreased in the cardiomyocytes co-cultured by the C-kit+ cellsas compared by control group.Conclusion: In this experiment, it can be concluded that changesinduced in the gene and protein expression level in cardiomyocytederived adult rat heart co-cultured with C-kit+ bonemarrow stem cells via VEGF signaling pathway.
