Down-regulation of the LAMB۳ gene induces pulmonary metastasis in thyroid cancer

The presence of distant metastases in thyroid cancer patients is only detected in ۱% to ۴% ofpatients with a poor prognosis. Metastasis sites for thyroid cancer are typically located in the lungs,bones, brain, and liver, with the lungs representing the most common metastatic site (۴۳%). The overallsurvival rate for patients with lung metastasis ranges from ۲۵% to ۷۵%. Laminin Subunit Beta ۳(LAMB۳) has been studied in lymph and liver metastasis, but no significant study has been conductedon thyroid tumor metastasis to lung tissue. In order to determine whether LAMB۳ affects pulmonarymetastasis, differentially expressed genes (DEGs) were identified from tissues with pulmonarymetastasis. Nine datasets were selected from the gene expression omnibus (GEO) databank. Each dataset was standardized using the Log۲ transformation and quantile normalization. A meta-data set wasconstructed, and the limma statistical package was used to identify DEGs. In order to adjust P-values,the Benjamini-Hochberg procedure was used. To assess the specificity of the amplification, RT-qPCRreactions will be performed, and melt curve analysis will be performed. The Livak method would beused to assess the relative expression of LAMB۳. Microarray meta-data analysis showed that theLAMB۳ gene was significantly downregulated (logFC = -۲.۴۲۹۳۱, adj.p.val = ۱.۰۲E-۰۸). In conclusion,pulmonary metastases are characterized by a low expression of the LAMB۳ gene, indicating that thisgene plays a tumor-suppressive role.
