The effect of in ovo serotonin injection on heart HspP۷۰ and Hsp۹۰ gene expression in heat stressed broiler embryo

Elevated environmental temperatures and heat stress adversely affect physiological performance. Chicks, lacking sweat glands, serve as a unique model for studying heat stress due to their heightened sensitivity, resulting in elevated body temperature and oxidative stress. Serotonin (۵-hydroxytryptamine, ۵-HT) plays a pivotal role in stress responses across species, influencing physiological and metabolic adaptation to adverse conditions. Heat shock proteins (HSPs) are a group of proteins synthesized in response to various stressors, safeguarding cells from damage. This study aimed to explore the impact of serotonin (۲۰ micrograms per egg) on the expression of HSP۷۰ and HSP۹۰ genes following exposure to high temperatures (۳۹.۵°C) for varying durations (۲, ۴, ۶, ۸, and ۱۰ hours). Analysis of data showed heat stress notably upregulated HSP۷۰ while downregulating HSP۹۰ gene expression compared to normal temperature conditions. Conversely, in ovo serotonin injection led to increased expression of both HSP۷۰ and HSP۹۰ genes, particularly under heat stress. This discovery in Ross broiler embryos highlights alterations in heart HSP۷۰ and HSP۹۰ gene expression under heat stress conditions, suggesting serotonin's role as an anti-stress agent inducing HSP gene expression. These findings propose the potential use of nutritional supplements in the poultry industry to regulate HSPs especially in heat stress condition
