This paper provides a comprehensive overview of protein-mediated covalent modifications appliedto Polycaprolactone (PCL) scaffolds, highlighting their transformative impact on tissue engineering anddrug delivery. The review synthesizes recent advancements, emphasizing tissue regeneration process andtargeted drug delivery achieved through the covalent attachment of bioactive proteins. Examining theimplications for tissue regeneration, success stories include bone, cartilage, vascular, and neuralapplications. The discussion extends to the versatility of modified PCL scaffolds in drug delivery,emphasizing precise temporal control and spatial targeting. Future perspectives underscore emergingtrends in research, encompassing diversification of proteins, innovative combination strategies, clinicaltranslation, and the development of dynamic, responsive systems. This review positions the integrationof protein-mediated covalent modifications and PCL scaffolds as a transformative approach withsignificant implications for the future of biomaterials in biomedical applications, promising personalizedand effective therapeutic interventions.
