Introduction: Chronic myeloid leukemia (CML), the first type of leukemia to be identified, remains fatal evenwith treatments such as stem cell transplantation, chemotherapy, and radiation therapy. Drug resistance andrecurrence remain problems, despite the potential of new approaches based on immunotherapy and cell therapy.Nanoparticles offer a potential solution because they reduce toxicity and side effects while enabling a drug tobe released gradually. Combining the chemotherapy medication doxorubicin (DOX) with the naturallyoccurring polyphenol Curcumin (CUR) in silver nanoparticles (AgNPs) is one possible strategy. In addition toaddressing the drawbacks of DOX, this combination promotes a gradual and continuous release of medication,which may result in a breakthrough in the safe and efficient treatment of CML.Methods: AgNPs were produced using a simple and fast chemical method. After that, to form a stable drugdelivery system. Formulations were prepared by loading DOX and CUR in AgNPs. The cytotoxic effects ofAgNPs were evaluated on Human Leukemia cell line (K۵۶۲). The drug release profile was also investigated inPBS (pH ۷.۴). In addition, the expression levels of BCL-۲ and MDR۱ mRNA in the K۵۶۲ cells treated with theAgNPs were assessed by Real-Time PCR.Results: In this study, our research focused on the synergistic effects of doxorubicin and curcumin encapsulatedin silver nanoparticles for the treatment of chronic myeloid leukemia (CML). The results demonstrated asignificant reduction in the growth of CML cells through the synergistic interaction of these compounds. Theutilization of silver nanoparticles not only mitigated the side effects of the drugs but also enhanced theirefficiency and intracellular density within leukemia cells. These findings highlight the potential of thisinnovative approach in improving the treatment of chronic myeloid leukemia while minimizing adverse effects.Conclusion: Silver nanoparticles exhibit immense potential for therapeutic application in treatment of leukemia.In addition to therapeutic uses, they also have shown a role in improving detection and diagnosis of leukemia.By incorporating both DOX and CUR into silver nanoparticles, a novel strategy to boost these medications'anticancer efficacy and prevent multidrug resistance in K۵۶۲ cells is presented. The promising findings of thiswork highlight how nanotechnology may completely transform cancer treatments, and they call for moreresearch to clarify the underlying mechanisms and improve the formulation for therapeutic use. Finally, thisstudy adds important knowledge to the creation of novel approaches to counter multidrug resistance in cancertreatment.
