BACKGROUND AND OBJECTIVESDue to the importance of microalgae as organisms that are used in many biological processes and the production of high value materials and recombinant products, their preservation has always been considered. Microalgal strains generally are maintained in laboratories through serial subculture.MATERIALS AND METHODSIn this research, some protocols have been used for the preservation of two microalgae strain, Spirulina platensis and Chlamydomonas reinhardtii. These protocols, consist of Lyophilization (Freeze-Drying), Cryopreservation at – ۲۰ to -۸۰° C and liquid nitrogen at under -۱۳۰ ° C , using different concentration of DMSO (dimethylsulfoxide), Glycerol , Methanol, ethylene glycol, sucrose and sorbitol. In addition to the usual methods of subculturing in agar medium has been used.RESULTS AND DISCUSSIONThe results showed that the Viability of microalgae by using these methodologies is different and it is evident that the microalgae can be preserved for a long time without using expensive equipment and liquid nitrogenCONCLUSIONThis technique requires a lot of human power and is often time-consuming and leads to genetic change. The present study is aimed to develop an alternative method for the long-term preservation of stock cultures.
