Microbial profiling of Paederus fuscipes (Coleoptera: Staphylinidae) through Next-Generation Sequencing

BACKGROUND AND OBJECTIVESPaederus fuscipes is the most famed beetle, which causes dermatitis or conjunctivitis in humans via discharging pederin toxin. Pedrin with antitumor and antiviral properties have been shown to be synthesized by uncultured Pseudomonas-like endosymbionts. This study was aimed to characterize the structure and diversity of P. fuscipes microial communities.MATERIALS AND METHODSAdult rove beetles were gathered from June–July ۲۰۱۹–۲۰۲۰ from the humid areas of Guilan, Mazandaran, and Golestan provinces where highest cases of linear dermatitis have been reported. The Illumina HiSeq sequencing platform was used to determine the bacterial diversity of the ۱۶S rRNA gene (V۳-V۴) in insect samples in terms of gender, organ, and location.RESULTS AND DISCUSSIONThe OTUs identified from P. fuscipes specimens were collapsed into ۴۰ phyla, ۱۱۲ classes, ۲۴۹ orders, ۳۶۵ families, ۵۷۶ genera, and ۱۰۶ species. Thirty top genera made up > ۹۴% of the P. fuscipes microbiome, with predominating Pseudomonas, followed by the Spiroplasma, Apibacter, Enterococcus, Dysgonomonas, Sebaldella, Ruminococcus, and Wolbachia. Spiroplasma / Apibacter as well as Pseudomonas / Pseudomonas were the most abundant in the genitals / intestines of male and female beetles, respectively. Additionally, male and female rove beetles were characterized by distinctive microbiota in different organs, likely reflecting different functions and/or adaptation processes.CONCLUSIONThese findings may eventually lead to ecological insights into the production and utilization of defensive compound of pederin as well as the management of linear dermatitis with the use of available antibiotics.
