Expression analysis of lncRNA LINC۰۰۲۶۱in tumor and marginal tissues of patients with colorectalcancer

چکیده مقاله:
Background: Colorectal cancer is the second most commoncause of cancer death worldwide. This cancer usually beginswith forming of small and benign masses called polyps in theinner part of the large intestine. The regulation of gene expressionis one of the fundamental processes that regulate all kindsof cellular mechanisms, and changes in some of these processescan cause cells to undergo uncontrolled proliferation. A widerange of molecules is involved in gene expression regulation,among which long non-coding RNA molecules (lncRNAs) playa significant role in gene expression regulation. Studies showthat lncRNAs can be involved in screening, diagnosis, prognosis,and treatment Results: Among these lncRNAs, the lncRNALINC۰۰۲۶۱ gene acts as a negative regulator of cell growth andcan promote differentiation and apoptosis.Materials and Methods: In this study, the expression levelof the lncRNA LINC۰۰۲۶۱ gene in ۴۷ patients with colorectalcancer was investigated using Real-Time PCR. The receiveroperating characteristic (ROC) curve analysis was performedto evaluate the potential of the prognostic biomarker in CRC.Results: Data analysis with SPSS and GraphPad Prism softwareillustrates decreased lncRNA LINC۰۰۲۶۱ gene expressionlevels in tumor tissues compared to tumor peripheral tissue (Pvalue=۰.۰۰۴). However, the low expression level of this genedid not show a significant relationship with clinicopathologicalsymptoms. In addition, the ROC curve was obtained with۶۸.۰۹% specificity,۶۵.۹۶% sensitivity, and ۰.۶۷ area under thecurve (AUC). Conclusion: Considering the significant decrease inLINC۰۰۲۶۱ gene expression in tumor tissue compared to thetumor margin tissue and the results obtained from examiningthe ROC curve, the gene LINC۰۰۲۶۱ can be considered one ofthe predictive and therapeutic biomarkers for colorectal cancer
