Background and Objectives: Semen quality and related parameters correlate directly with fertilization and consequently assisted reproductive technology outcome. Traditional studies on carob (Ceratonia siliqua) have demonstrated its effect on male fertility potential via reductive effect on reactive oxygen species. The antioxidant effect of carob kibble extract on sperm motility characteristics in cryopreserved semen samples was investigated in this study. Material and Methods: The extract was made using acetone as solvent followed by vacuum evaporation and filtration. Following thawing, each of forty human semen samples were divided into four groups and exposed to concentrations of ۰.۰ (control), ۰.۰۵, ۰.۲, and ۰.۸ mg/ml of the extract. Percentages of progressive motile, non-progressive motile and immotile sperms as well as other kinematic parameters were assessed by computer aided sperm analysis, immediately after exposing to the concentrations (T۰) and one hour later (T۱). Data was analyzed by repeated measure analysis of variance and paired sample t-student tests using SPSS software. Level of p< ۰.۰۵ was considered statistically significant.Results: There was no significant difference between groups in all values at T۰ or T۱. However, comparison of matched doses at T۰ and T۱ indicated that lower doses ۰.۰۵ and ۰.۲ mg/ml could significantly (p<۰.۰۵) rule out natural decline in motility.Conclusion: Although adding lower doses of carob kibble extract on thawing medium could have supportive effect on sperm motility, however adding extract to vitrification solution before freeze-thawing process as well as orally intake of the extract seem to have more efficiency that would be a subject for further studies.
