Objective: Vimentin is an intermediate filament (IF) that is oneof the firs t filaments to be expressed during spermatogenesis.Vimentin has a variety of functions, including cell shape determination,differentiation, cell motility, cell junction maintenance,intracellular trafficking, and helping in the preservationof proper differentiating of germ cell morphology.Materials and Methods: This s tudy focused at vimentin expressionin two groups of spermatogonia: undifferentiated anddifferentiated. We used immunocytochemis try (ICC), immunohistochemis try (IHC), and Fluidigm real-time polymerase chainreaction (PCR) to evaluate the vimentin expression in vivo andin vitro.Results: According to IHC data, s trong vimentin expressionwas found in the center of seminiferous tubules, whereas lowexpression was seen in the basal membrane. Under in vitro conditions,ICC examination of colonies by isolating differentiatedspermatogonia revealed positive expression for the vimentinantibody, whereas vimentin expression was negative in theundifferentiated population. Fluidigm real-time PCR analysisrevealed that vimentin expression was significantly higher indifferentiated spermatogonia than in undifferentiated spermatogonia(P value <۰.۰۵). Our findings revealed that vimentin waselevated during the spermatogenesis differentiation phases,demons trating that vimentin is an intermediate filament withcritical involvement in the differentiation s tages of tes ticulargerm cells.Conclusion: These findings promote further research into thespermatogenic process, both in vitro and in vivo
