Screening of Different Eryngium Extracts on BetaCell Regeneration Using Transgenic Zebrafish Model

Objective: In type ۱ diabetes and at the final s tages of type ۲ diabetes,pancreatic beta cells mass is decreased. One of the proposedapproaches for beta cells replacement is endogenous regenerationof these cells. Zebrafish is a powerful model for drugscreening s tudies, because of regenerative capacity in differentorgans, transparency in larvae s tage, and high fecundity. Manys tudies have been conducted to find the effective compound inregeneration of beta cells that have been led to identificationof different signaling pathways and small molecule inducers.Iranian traditional medicine has advised some medical plantfor diabetes treatment including Boghnagh (genus Eryngium)which its biologically functional compounds and mechanismsof action needs further inves tigation.Materials and Methods: In this s tudy, different species ofBoghnagh plant was collected from different regions in Iran andextracts were obtained from root, leave, and aerial parts of thisplant. The larvae of transgenic zebrafish Tg(ins:CFP-NTR) at ۳days pos t fertilization was exposed to metronidazole (MTZ) forablation of beta cells. After ۲۴ hours and washing of MTZ, thelarvae was treated with different concentrations of extracts for۲ days. The effect of treatments on beta cell regeneration wasevaluated by imaging of larvae under fluorescent s tereomicroscope and measuring the beta cell area by imageJ software.Results: Results showed that the ethyl acetate extracts fromaerial parts gathered from special regions of the country, havesignificant effects (P value <۰.۰۵) in beta cell regeneration incomparison with controls groups.Conclusion: These findings can led us to further fractionations tudies of the extract to find the hit compound/compounds andalso for further pre-clinical s tudies using higher animals of betacell regeneration
