Leucine and Endurance Exercise Altered The HubGenes and LncRNAs Networks in Brain-Gut Axis of Micewith Depression Like Behaviors

Objective: In developing and developed countries, psychologicaldisorders have imposed heavy economic and social burdenson health care sys tems. Depression is a frequent mood disorderthat might impair the brain-gut axis.Materials and Methods: Artificial intelligence indicated amRNAs-lncRNAs network involved in the depression condition.In-silicon and chemoinformatics analysis revealed thatfour genes, VEGF-α, KDR, PTEN, and BDNF, with the highes tdegree and mos t betweenness, could be affected by protein-proteininteraction networks under influencing endurance exerciseand leucine consumption. We divided ۳۰ mice into five groups:untreated mice, mice with depression-like behaviors, mice withdepression-like behaviors treated with consumed leucine, micewith depression-like behaviors treated with exercise training,and mice with depression-like behaviors treated with exercisetraining along with consumed leucine.Results: According to artificial intelligence biological analysis,we found some mediators such as lncRNAs profile and Kdr/Vegf-α/Pten/Bdnf interaction network in the hippocampus regionand ileum tissue which could be decisive molecules in thebrain-gut axis. Moreover, KDR as a principal cutpoint proteinin the network was identified as the pharmaceutical approachfor major depressive ameliorating based on pharmacophoremodeling and molecular docking outcomes. Furthermore, weindicated that the mRNA level of the Pten enhanced and Vegf-α/Kdr/Bdnf mRNAs decreased in mice with depression-like behaviors.Moreover, exercise and leucine ameliorated the braingutaxis in mice with depression-like behaviors.Conclusion: Exercise and leucine regulated the lncRNAs network in the hippocampus and ileum of mice with depressionlikebehaviors. We sugges t that the lncRNAs profiles couldconsider as diagnosis and prognosis biomarkers, and exercise+ leucine might be a practical approach to improve depression.
