Sequence analysis of the flaB۲ gene among Leptospirainterrogans serovars from Iran

چکیده مقاله:

Background and Aim : Leptospira interrogans is a bacteria belonging to the leptospiraceae familythat can cause a common infection in humans and animals by pathogenic Leptospira. Severalfactors are known as virulence factors of L. interrogans such as flagellins, adhesions, outermembrane proteins. Motility is an essential virulence factor for the pathogenic species thatencoding flagellum protein. The aim of this study was to analyse the nucleotide sequence of flaB۲gene L. interrogans from Iran and phylogenetic comparison with isolates from other countries.Methods : In this study, ۱۲ pathogenic Leptospira serovars and two non-pathogenic Leptospiraserovars were used. The pathogenic serovars of Leptospira interrogans were cultured into theselective medium EMJH and the genomic DNA was extracted by standard Phenol-Chlorophormmethod. The flaB۲ gene were amplified with PCR and specific primers. The nucleotide sequencesof flaB۲ gene were analysed by MegAlign software with NCBI database.Results : The PCR product of flaB۲ gene was ۱۰۵۰ bp. All pathogenic serovars of L. interroganscontained flaB۲ gene. No PCR products were amplified from two non-pathogenic L.biflexa.Sequence analysis by BLAST showed ۱۰۰% similarity among similar pathogenic serovarscompare with published sequences in the GeneBank.Conclusion : The results of this study showed that the flaB۲ gene was highly conserved amongpathogenic Leptospira. The results suggested that may useful for diagnostic of leptospirosis orcandidate for recombinant vaccine and can also be used in an ELISA kit for the serodiagnosis.
