Backgrounds: Prostate cancer is the commonest cancer in men, affecting ۲۱۴ per ۱۰۰۰European men. It is the second commonest cause of cancer death. Estrogens play a role inproliferation in the prostate, but interestingly are capable of stimulating as well as inhibitinggrowth. Recent studies have demonstrated that estrogen receptor signaling pathways may beimportant in the development and maintenance of BPH and Prostate cancer. The aim of thisstudy is to investigate the effect of estrogen on BAX gene expressions in Prostate Cancer-۳ (PC۳)cell lines.Materials and Methods: In this study, prostate cancer cells of the PC۳ cell line were cultured inDMEM medium with ۱۰% bovine serum. Cells were treated at ۲۴, ۴۸ and ۷۲ hours’ intervals.The cells were exposed to concentrations of ۱۰, ۲۰, ۳۰, ۴۰, ۵۰, ۶۰, ۷۰, ۸۰, ۹۰, ۱۰۰ nμ ofEstrogen hormone and the cell growth was measured by MTT colorimetric method. Thenexpressions of p۵۳ and BAX genes in cells exposed to ۱۰۰ nm Estrogen hormone concentrationswere investigated by real-time PCRResults: By increasing the concentration of Estrogen hormone, and time-dependently, whileincreasing the induction of apoptosis, the bioavailability of the cells and the expression level ofBAX gene decreased. The maximum Effect was gained by ۱۰۰ nm of Estrogen hormone and ۷۲hours after treatment of the cells.Conclusion: The prostate is an estrogen target tissue and estrogens directly and indirectly affectgrowth and differentiation of prostate.
