Backgrounds: Benign Prostatic Hyperplasia and Prostate cancer are common clinicalcomplications in urology. Estrogen receptor signaling pathways may be important in thedevelopment and maintenance of BPH and Prostate cancer. Estrogen and Selective EstrogenReceptor Modulators (SERMs) have shown a potential role in promoting or inhibiting prostateproliferation in Prostate cancer. The aim of this study is to investigate the effect of estrogen onDLGAP۵ gene expressions in Prostate Cancer-۳ (PC۳) cell lines.Materials and Methods: In this study, prostate cancer cells of the PC۳ cell line were cultured inDMEM medium with ۱۰% bovine serum and treated at ۲۴, ۴۸ and ۷۲ hours’ intervals. The cellswere exposed to concentrations of ۶۰, ۸۰, ۱۰۰ nμ of Estrogen hormone. Apoptosis and cellgrowth were assessed by flow cytometry with Annexin-PI kit according to the kit instructions.Then expressions of DLGAP۵ gene in cells exposed to ۱۰۰ nμ Estrogen hormone concentrationwere investigated by Real Time RT-PCR.Results: By increasing the concentration of Estrogen hormone, and time-dependently, whileincreasing the induction of apoptosis, the expression level of DLGAP۵ gene decreased. Themaximum Effect was gained by ۱۰۰ nμ of Estrogen hormone and ۷۲ hours after treatment of thecells.Conclusion: Estrogen hormone, in a dose-dependent and time-dependent manner, decreases cellgrowth, increases induction of apoptosis and reduces the expression of these genes examined inPC۳ cells. According to the current evidence, estrogen hormone can act as an anti-cancer agent.
