Backgrounds: We examined four single nucleotide polymorphisms in four antioxidant genes(PON۱, CAT, GPx۱, and SOD۲) in ۱۰۰ infertility cases and ۱۰۰ controls from an Iranianpopulation-based case-control study to confirm the assumption that polymorphisms in oxidativestress genes increase the risk of sperm DNA damage and idiopathic male infertility.Materials and Methods: Restriction fragment length polymorphism and tetra-primeramplification refractory mutation system PCR were used to identify genotypes. Sperm DNAdamage was assessed using the Sperm Chromatin Dispersion test (Halo Sperm), and the totalantioxidant capacity of seminal fluid was determined using the FRAP assay.Results: Our findings demonstrated that alleles Arg-PON۱ (rs۶۶۲) and Ala-MnSOD (rs۴۸۸۰)variant genotypes were considerably linked with a higher risk of male infertility.Conclusion: Linear regression analysis revealed that those with the PON۱ Gln۱۹۲Arg or SOD۲Val۱۶Ala variants have significantly higher levels of sperm DNA fragmentation and lower levelsof the total antioxidant capacity in seminal fluid.
