Investigating differential expression of mTOR۱/UCA۱ in tumor samples of colorectal cancer compared with tumor marginal samples

Backgrounds: Colorectal cancer (CRC) is the second and third most common cancer in men andwomen respectively, and the fourth cause of cancer death of individuals. Mutations in specificgenes can lead to colorectal cancer. UCA۱ is one of the oncogenic genes that have been shown tostimulate cell proliferation. mTOR۱ is another gene that leads to the growth of cancer cellsthrough anabolic processes and autophagy inhibition. In this study, we evaluate the expression ofthese two genes in different phases of CRC that helps the early detection of colorectal cancerwhich can increase the survival rate.Materials and Methods: First, we collected ۲۵ colorectal cancer tumor tissues and ۲۵ adjacentnormal tissues as a control group. Then, RNA was extracted from tissue samples and cDNAsynthesized. The UCA۱ and mTOR۱ expression was evaluated in CRC tissues compared toadjacent normal tissues by Real Time PCR after synthesizing specific gene primers.Results: Our results showed that the UCA۱ and mTOR۱ expression in the tumor tissues wassignificantly higher than in the adjacent normal tissues (P <۰.۰۵). There was also a significantdifference in Lynph inv and Vescu inv with mTOR۱ expression (p <۰.۰۵).Conclusion: Our results showed that UCA۱/mTOR۱ may be important genes involved incolorectal cancer. mTOR۱ was also identified as one of the possible genes in metastasis ofcolorectal cancer. Thus, UCA۱ and mTOR۱ can probably be considered as biomarkers in CRCtherapy and diagnosis.
