Backgrounds: Hepatocellular carcinoma (HCC) is the most common primary liver cancer and isa serious medical problem, as its incidence is continuing to grow. Nuclear factor erythroid ۲-related factor (Nrf۲) is an important transcription factor associated with HCC carcinogenesis,and increased NRF۲ mRNA levels contribute to the poor patient outcome for HCC. Here, theeffect of Nrf۲ knockdown by siRNA was examined in the HCC cell line.Materials and Methods: First, because of the high expression level of Nrf۲, the hepG۲ cell linewas chosen for further research. Subsequently, the cells were cultured in a ۱۰% RPMI mediumto reach ۷۰% confluence. They were then transfected with Nrf۲-specific siRNA and theexpression level of Nrf۲ was evaluated by qRT-PCR. After that, changes in cell viability andapoptosis were analyzed by MTT assay and annexin V-FITC/PI staining respectively.Results: Nrf۲-specific siRNA significantly suppressed the Nrf۲ expression, decreased the cellviability, and promoted the hepG۲ cells apoptosis.Conclusion: Overall, the results of this study showed that Nrf۲ knockdown might be consideredas a potential approach for Hepatocellular carcinoma management.
