Study of the relationship between FLT۳, hsa-miR-۱۵۰-۵p, and the progression of malignancy in patients with leukemia

چکیده مقاله:

Introduction: Leukemia is a cancer of the hematopoietic tissues of the body, including the bone marrow and lymphatic system. The tyrosine kinase ۳ (FLT۳) gene, which encodes a membrane-bound receptor tyrosine kinase, plays an essential role in normal hematopoiesis. FLT۳ mutations are considered high-risk factors in leukemia. This gene is overexpressed in leukemia and patients with high expression of FLT۳ simultaneously express high levels of leukemic oncogenes. Therefore, high expression of FLT۳ is a risk factor in leukemia. FLT۳ gene is specifically overexpressed in acute lymphoid and myeloid leukemia (ALL and AML) (۱). A large number of MicroRNAs are tumorigenic, while others have tumor suppressor effects. Previous studies showed that the expression of miR-۱۵۰ is decreased in patients with AML, and this miRNA acts as a tumor suppressor in AML patients (۲).Material and methods: First, using the Enrichr site, the FLT۳ gene was selected as a gene involved in leukemia. FLT۳ was selected as the target gene for the hsa-miR-۱۵۰-۵p by the miRTarBase section of this site. By selecting FLT۳ as the target gene and hsa-miR-۱۵۰-۵p as the miRNA, the results of their interaction were investigated in the miRwalk database. The findings related to the interaction of these two sections can be observed in the miRTarBase.Results: According to the findings of the miRTarBase site, hsa-miR-۱۵۰-۵p by targeting the ۳'UTR region of the FLT۳ gene and with the help of the RISC complex leads to the suppression of the mRNA translation of the target gene. hsa-miR-۱۵۰-۵p sequence binds to nucleotides ۱۶ to ۳۷ of the target mRNA gene.Conclusion: According to previous studies, bioinformatic predictions, the oncogenic role of the FLT۳ gene, and the function of the microRNAs negative regulation, it is expected that the expression of the hsa-miR-۱۵۰-۵p will decrease, and then, the expression of FLT۳ gene will increase. For this reason, it is predicted that hsa-miR-۱۵۰-۵p has an anti-tumor function and the decrease in the expression of this miRNA leads to an increase in the expression of the FLT۳ gene, tumor growth, and the development of malignancy in leukemia.
