Protective Effects of Coenzyme Q۱۰ on antioxidant capacity and lipid peroxidation in solvents-exposed printing workers

Background and Objective: The efficacy of CoQ۱۰, as a well-tolerated and safe supplementation, in protecting the oxidative damage caused by occupational exposure to organic solvents is questionable The study aimed to determine whether coenzyme Q۱۰ reduces oxidative stress induced by toluene and xylene in printing workers who were occupationally co-exposed to these solvents.Materials and Methods: In a randomized, double-blind, placebo-controlled ۸-weeks crossover trial, coenzyme Q۱۰ (۶۰ mg twice daily) or placebo was administrated to ۴۰ printing workers occupationally exposed to mean levels of hippuric acid (۰.۱۵ ± ۰.۰۴ g g−۱ creatinine) as well as ortho- (۰.۱۱ ± ۰.۰۱ g g−۱ creatinine), and meta- ¶ methyl hippuric acids (۰.۱۹± ۰.۰۷ g g−۱ creatinine) as the urinary biomarkers of exposure for toluene and xylene isomers respectively, in printing industry. The subjects were randomly assigned to the placebo (n = ۲۰) and coenzyme Q۱۰ groups (n = ۲۰). The serum levels of total antioxidant capacity (TAC) and malondialdehyde (MDA) as the marker of lipid peroxidation were measured pre- and posttreatment phases. The outcome variables were ۸-week changes in mean the serum levels of TAC and MDA from baseline between coenzyme Q۱۰ and placebo groups. The study protocol was approved by the Ethics Committee for Medical Research (No. IR.ZAUMS.REC.۸۵.۵-June-۲۰۱۶) and registered with the Iranian Registry of Clinical Trials (No.IRCT۲۰۱۶۰۶۱۲۲۸۴۰۷N۱).Results: The results of this study showed that, compared with the placebo, ۸-weeks of coenzyme Q۱۰ supplementation was associated with significantly reduction in serum MDA levels (F (۱, ۳۸) = ۷۵.۳۸, P<۰.۰۰۱). In the CoQ۱۰ group, the MDA levels were significantly lower than baseline after ۸-weeks (۵.۶۸±۱.۳۳ vs ۷.۰۷±۱.۷۷ µmol. L-۱, P<۰.۰۵). In addition, there was a significant increase in the mean serum TAC levels of printing workers following coenzyme Q۱۰ administration compared with placebo (F (۱, ۳۸) = ۵.۰۶, P=۰.۰۳).Conclusion: The study indicated that Coenzyme Q۱۰ at a dose of ۶۰ mg twice daily may induce protective effect in printing workers, resulting in reduced lipid peroxidation andincreased antioxidant capacity. Further confirmation is needed to verify these observations with a larger number of subjects.
