The Effects of ۸ weeks of HIIT and N-Chromosomes Royal Jelly on the Expression of NKX۲.۵ and GATA۴ Genes in the Heart Tissue of Type ۲ Male Diabetic Rats

Cardiovascular diseases are the main cause of death in diabetic patients. Exercise and antioxidants are known to have a protective effect against the risk of diabetic heart by reducing the effects of free radicals. the purpose of current study was to investigate the effects of ۸ weeks of high-intensity interval training (HIIT) and N-chromosomes royal jelly on the expression of Nkx۲.۵ and Gata۴ genes in the heart tissue of male type ۲ diabetic rats.This experimental study was performed on ۳۶ male diabetic rats were randomly divided into four groups: control (n=۸), HIIT (n=۱۰), N-chromosomes royal jelly (n=۸) and HIIT*N-chromosomes royal jelly (n=۱۰) groups. The HIIT training intervention of ۸ weeks, including ۲ to ۸ intervals with ۲ min running at ۸۰-۹۰% VO۲max and ۱ min at ۵۰-۵۶% VO۲max, was performed for ۵ sessions per week. Additionally,۱۰۰ mg/kg N-chromosomes royal jelly consumed, ۵ days/week, in supplement groups. Both Nkx۲.۵ and Gata۴ genes expression in heart tissue was measured by real-time RT-PCR. To evaluate the differences between the groups, ANOVA and LSD post hoc test were used at the significance level of p ≤ ۰.۰۵Analysis revealed that expression of Nkx۲.۵ gene was significantly increased in N-chromosomes royal jelly group and also in HIIT*N-chromosomes royal jelly group when compared with control group. Expression of Gata۴ gene was significantly increased in HIIT*N-chromosomes royal jelly group compared to the control group. whereas expression of Gata۴ gene was non-significantly increased in other intervention groups compared to the control group (p>۰.۰۵).HIIT and consumption of royal jelly by increasing markers of proliferation and differentiation of cardiomyocytes such as Nkx۲.۵ and Gata۴ genes causes cardiac growth and improvement of cardiac function in diabetic rats, probably interaction of HIIT and consumptions N-chromosomes royal jelly in diabetic patients is an effective non-pharmacological strategy.
