Metabotropic Glutamate receptor ۳ (mGlu۳) is a Receptor for glutamate that functions as ligand-gated ionchannel in the central nervous system and plays an important role in excitatory synaptic transmission. Thisreceptor is a type of multi-pass membrane protein. mGlu۳ is one of the therapeutic targets due to its geneticassociation with psychiatric disorders. It seems that the use of protein structure modeling and predictionmethods on Glutamate receptor ۳ protein with the aim of developing more effective and efficient agonistsfor the treatment, can have a significant effect on promoting Drug Discovery studies. For this purpose, thestructure of the protein predicted by I-TASSER, Robetta and SWISS-MODEL server. The best modelselected and subjected to ProSA and VADAR servers in order to determine the quality of the predictedstructure. The Z-score of the model was -۹.۳۱ and the percentage of amino acids in most favored regions was۸۸.۱۶۱. Then the protein structure was placed into a membrane model by CHARM-GUI server. Overally thepredicted model could be an ideal structure for further studies such as docking. In this regard the result ofmolecular docking studies using the predicted model as receptor and L-glutamate as ligand shows that thebest binding pose has -۳.۵۸ kCal/mol.
