Effects of an oligosaccharide isolated from Rosa canina on sensory-motor function in a clip compression model of spinal cord injury

Background and Objective Spinal cord injury (SCI) causes several sensory-motor and neurological complications followed by secondary events such as inability to move an organ, loss of sexual function, decreased ability to control urination and defecation, as well as excruciating pain occur. There is currently no effective treatment for post-SCI complications. Therefore, conducting research to find new therapeutic goals in these patients is of great importance. Considering the critical role of inflammation and apoptosis in the pathogenesis of SCI, as well as the anti-inflammatory and anti-apoptosis property of oligosaccharides in neurodegenerative diseases, we decided to investigate the effects of Rosa canina oligosaccharide (RCO) on improving sensory and motor function post-SCI.Materials and Methods Thirrty-five adult male rats were kept in standard laboratory conditions and randomly divided into five equal groups called sham, SCI, and oligosaccharide treatment with ۵, ۱۰, and ۱۵ mg RCO/ml normal saline. After laminectomy, spinal cord injury was performed by compression method using an aneurysm clips. Behavioral tests of hot plate, acetone drop, inclined plane, BBB as well as weight measurement were performed on days ۷, ۱۴, ۲۱ and ۲۸. In addition, the activity level of antioxidant enzymes was checked.Findings RCO in ۵mg/ml dose reduced mechanical and thermal pain in the studied rats and also improved the use of joints and limbs in motor tests. According to the molecular results, the activity level of antioxidant enzymes increased significantly compared to control groups.ConclusionThe results suggest RCO as a promising candidate to combat against neuropathic pain and some other major complications, through the suppression of oxidative pathways, post-SCI.
