Antiproliferative effects of fresh water crab hemolymph and meat extract on breast cancer cell line

Background and aims: Despite the advances in drugs, side effects of chemotherapy drugs continue to exist. Therefore, more attention hasbeen paid to the compounds derived from medicinal herbs and aquatic organisms. This study aimed to investigate the effect of freshwatercrab hemolymph and meat extract on breast cancer (BC) cell line (۴T۱).Methods: After isolation of freshwater crab hemolymph and meat extract, protein concentration and total antioxidant capacity wereanalyzed by bicinchoninic acid (BCA) and cupric reducing antioxidant capacity (CUPRAC) methods. The ۴T۱ cells and bone marrowmesenchymal stem cells (BMSCs) were treated with crab hemolymph (۱, ۲, ۱۰ mg/mL) and meat extract (۰.۱, ۰.۲ and ۱ mg/mL), and cellsurvival was analyzed using ۳-(۴, ۵-dimethylthiazol-۲-yl)-۲,۵-diphenyltetrazoliumbromide assay (MTT) at ۴۸ and ۷۲ hours. Nitric oxide(NO) secretion was measured by Griess method. Data were analyzed using one-way analysis of variance (ANOVA).Results: Protein concentration of ۲۳.۲۵ mg/mL was shown in crab hemolymph, and ۲.۳ mg/mL in meat extract. Total antioxidant capacitywas reported as ۱.۰۳۶ μM/mL and ۱.۱۰۴ μM/mL in crab hemolymph and meat extract, respectively. Cell survival in the ۴T۱ cells wasdecreased in a dose- and time-dependent manner (P ≤ ۰.۰۰۱). NO secretion of ۴T۱ cells was decreased after treatment with differentconcentrations of crab hemolymph and meat extract at ۴۸ and ۷۲ hours. Cellular growth was observed in BMSCs after treatment withdifferent concentrations of crab hemolymph and meat extract at ۴۸ and ۷۲ hours.Conclusion: Since crab hemolymph and meat extract have protein and antioxidant activities, they can have anti-cancer effects on ۴T۱ cells.
