Differentiation-inducing activity of Epigallocatechin-۳-gallate (EGCG) and Kaempferol on NB۴ cells

The survival rate of patients with acute promyelocytic leukemia (APL) can be dramatically improved upon the treatment with all trans-retinoic acid (ATRA). However, the toxicity of this drug is a major problem in the treatment of APL. Previous studies have reported that phyto-polyphenols such as epigallocatechin gallate (EGCG) and kaempferol cause cell death in hematopoietic neoplasms without negatively impacting healthy cells. Methods: In the present study, the differentiation-inducing activity of EGCG and kaempferol was examined by NBT assay and Real-time PCR in leukemia NB۴ cells.Results: EGCG (۲۵ µM) and kaempferol (۵۰ µM) induced the differentiation of NB۴ cells towards granulocytic pattern in a similar manner observed for ATRA (۱ μM). Furthermore, EGCG suppressed the expression of clinical marker PML/RARα further compared to kaempferol, while also decreasing the expression of HDAC۱ in leukemia NB۴ cells.Conclusion: Based on these results, compared with kaempferol, EGCG at low concentrations merits more attention for long-term ATRA therapy in patients with APL.
