Study of human TNF-α -۳۰۸ (G/A) Gene Promoter Polymorphism in patients with Psoriasis using ARMS-PCR technique

Psoriasis is a common and reversible chronic immune disease of the skin and joints. genetic factors play an important role in the inflammatory and immune effects of psoriasis. One of the candidate genes involved in this disease is the TNF-α gene located on chromosome ۶ (۶p۲۱.۳) and encodes a multifunctional proinflammatory cytokine that is dependent on the tumor necrosis factor superfamily and is usually secreted by macrophages. There are several single nucleotide polymorphisms (SNPs) in this gene, the two most commonly polymorphisms being the conversion of G to A at the -۳۰۸- and ۲۳۸-position promoters, which are both functional and important. The aim of this study was to compare the frequency of rs۱۸۰۰۶۲۹ (-۳۰۸) G/A TNF-α gene polymorphism in psoriasis patients and healthy controls. This study was a case-control study performed on ۵۰ patients and ۳۰ healthy individuals in west Mazandaran and east Guilan. After extracting DNA of blood samples, (-۳۰۸) G/A TNF-α gene polymorphisms was studied using ARMS PCR method. The results were validated using DNA sequencing method. Data analysis were performed with SPSS۲۲ software. GG genotype and G allele were evaluated in patients with psoriasis. The frequency and percentage of GG genotype in control and patient groups were ۳۰ (۱۰۰%) and ۵۰ (۱۰۰%), respectively. The frequency and percentage of G allele in control and patient groups were ۶۰ (۱۰۰%) and ۱۰۰ (۱۰۰%), respectively (P <۰.۰۴۷). According to the results of TNF-α G/A polymorphism frequency in this study, there is no relationship between this polymorphism and susceptibility to psoriasis.
