Study of human Interleukin-۱۰ -۱۰۸۲(A/G) Gene Promoter Polymorphism in patients with Psoriasis using ARMS-PCR technique

Psoriasis is a chronic inflammatory skin disease that is prevalent among ethnic groups. IL-۱۰ is a cytokine produced primarily by monocytes and to be a lesser extent by lymphocytes. This gene is located on the long arms of chromosome number one (۱q۳۲.۱). Gene promoter Zone of it is very polymorphic and has three point mutations; also, rs۱۸۰۰۸۹۶ is considered in position of A/G (-۱۰۸۲). In general, this study compares the frequency of gene polymorphism rs۱۸۰۰۸۹۶ (۰۱۰۸۲) A/G IL-۱۰ in people with Psoriasis and healthy persons. This case-control study was a performed on ۵۰ patients and ۳۵ health individuals in west Mazandaran and east Guilan. After extracting DNA of blood samples, (-۱۰۸۲) A/G (rs۱۸۰۰۸۹۶) IL-۱۰ gene polymorphisms was studied using ARMS PCR method. The results were validated using DNA sequencing method. Data analysis were performed with SPSS۲۲ software. A frequency percentage of genotypes AG, GG and AG in the control group are ۰, ۱۸ and ۸۲ percent; and also ۵۶, ۳۲ and ۱۲ percent in the patient group, respectively. However, this difference in level ۰.۰۵ (p=۰.۰۱۴) is significant. Also, the frequency of alleles A and G in the control group are ۹ and ۹۱ percent; but in the patient group are ۷۲ and ۲۸ percent, respectively. So, this difference is significant in level ۰.۰۵ (p=۰.۰۰۲). Based on the results, it can be said that IL-۱۰A/G gene polymorphism is likely to play an important role in causing psoriasis.
