The major histocompatibility complex (MHC) in cattle is known as bovine lymphocyte antigen (BoLA). BoLA genes are organized into I, II, and III classes. At present, only one class I locus (BoLA-A) is internationally accepted on the basis of serological testing.DRB۳ is one of the class II genes which cattle express per haplotype. In current study, blood sampling was carry out from Holstein cattle (n=۳۰ susceptible and n=۳۰ resistant to mastitis) and a random sampling was conducted from Iranian native cattle (Sarabi; n=۳۰). DNA was extracted and amplified exon ۲ of BoLA-DRB۳ and BoLA-A genes using specific primers by PCR. PCR products were represented a ۲۸۴ bp and ۲۷۶ bp fragments on ۲% agarose gel respectively for DRB۳and BoLA-A. RFLP technique using Taq۱ endonuclease showed two genotypes, seven and five haplotypes respectively in Holstein and Sarabi breeds in DRB۳.۲ region. BoLA-A was monomorphic in both studied breeds either RFLP technique. This is the first study on characterization of exon ۲ of BoLA-A gene using RFLP techniques in Holstein and Sarabi breeds.
